متن کاملSelf-oligomerization regulates stability of survival motor neuron protein isoforms by sequestering an SCFSlmb degron.
Spinal muscular atrophy (SMA) is caused by homozygous mutations in human SMN1 Expression of a duplicate gene (SMN2) primarily results in skipping of exon 7 and production of an unstable protein isoform, SMNΔ7. Although SMN2 exon skipping is the principal contributor to SMA severity, mechanisms governing stability of survival motor neuron (SMN) isoforms are poorly understood. We used a Drosophil...
متن کاملA photosensitive degron enables acute light-induced protein degradation in the nervous system
Acutely inducing degradation enables studying the function of essential proteins. Available techniques target proteins post-translationally, via ubiquitin or by fusing destabilizing domains (degrons), and in some cases degradation is controllable by small molecules. Yet, they are comparably slow, possibly inducing compensatory changes, and do not allow localized protein depletion. The photosens...
متن کامل“ Photo ” Chemistry Without Light ?
In the early seventies, Giuseppe Cilento (São Paulo University), Emil White (Johns Hopkins University) and Angelo Lamola (AT&T Bell Laboratories) postulated that typical photochemical reactions could occur in dark parts of living organisms if coupled to enzymatic sources of electronically excited products. Their paradoxical hypothesis of “photochemistry without light” was chemically anchored on...
متن کاملGeneral method for regulating protein stability with light.
Post-translational regulation of protein abundance in cells is a powerful tool for studying protein function. Here, we describe a novel genetically encoded protein domain that is degraded upon exposure to nontoxic blue light. We demonstrate that fusion proteins containing this domain are rapidly degraded in cultured cells and in zebrafish upon illumination.
